Stretch-Activated Ion Channels in Stretch-Induced ANP Secretion

|
Mechanical stretch is the most important stimuli for the secretion of atrial ANP. However, the exact mechanism involved in the mechanosignal transduction is still unknown. The aim of the present study is to define the possible role of stretch-activated ion channels (SACs) in the ANP secretion using isolated perfused beating rat atria. An increase in atrial workload was induced by adjusting the height or the diameter of outflow catheter connected to perfused atrium. Both methods caused increases in pulse pressure similarly. Increasing catether height markedly increased ECF translocation and ANP secretion but decreasing the diameter slightly increased them. Changes in ANP secretion positively correlated to workload (loaded volume to atrium and change in pulse pressure). Inhibition of SACs by addition of gadolinium (0.1 mM) markedly attenuated stretch- induced increases in pulse pressure, ECF translocation and ANP secretion. The relation between changes in ANP secretion and workload was not observed by gadolinium. However, gadolinium did not affect changes in pulse pressure and ANP secretion induced by decreasing catheter diameter (which did not cause atrial stretch).
